CIA Induction Kits - For induction of collagen-induced arthritis (CIA) in DBA/1 mice

 

 

 

CIA is induced by chicken or bovine type II collagen in CFA, followed by a type II collagen booster 18 to 21 days later. The collagen booster is usually administered in IFA. Alternatively, the booster can be administered in CFA, resulting in faster CIA development and increased severity.

1. Consistently induces arthritis in 90 to 100% of DBA/1 mice (Taconic DBA1BO)
When used with the recommended protocol, 90 to 100% of mice develop arthritis within 35 days of immunization. Most mice reach disease severity of 10 to 14 (on the scale 0 to 16).
Each lot is tested and individually adjusted to ensure consistent disease induction.

2. Eliminate tedious preparation of emulsions
Properly prepared emulsions are critical for reliable induction of CIA. Our emulsions are carefully made and pre-filled into syringes, ready to use, to reduce time needed to set up experiments.

3. Save time and mice in testing individual reagents

Collagen-induced arthritis (CIA) is one of the most commonly used animal models of human rheumatoid arthritis (RA).

Joint inflammation, which develops in animals with CIA, strongly resembles inflammation observed in patients with RA. Blocking tumor necrosis factor (TNF) is efficacious in treatment of CIA, just as it is a highly efficacious therapy in treatment of RA patients.

CIA is mediated by both T cells and antibodies (B cells). Macrophages are believed to play an important role in mediating tissue damage during disease development.

CIA is induced by immunization with type II collagen/CFA emulsion, followed 18 to 21 days later by collagen/IFA or collagen/CFA emulsion.

While both bovine type II and chicken type II collagen are used for CIA induction, bovine collagen appears to be slightly better at inducing CIA in male DBA/1 mice, while chicken collagen works slightly better in females.

Protocol: CIA Induction in DBA/1 Mice

Data are from three independent experiments using Hooke Kits™ Chicken Collagen/CFA Emulsion (EK-0210) and Chicken Collagen/IFA Emulsion (EK-0211), with female DBA/1 mice (Taconic Biosciences).

 

 

Similar results are obtained using Bovine Collagen/CFA Emulsion (EK-0220) and Bovine Collagen/IFA Emulsion (EK-0221) in male DBA/mice (Taconic Biosciences).

For stronger CIA with earlier onset, use a booster dose containing collagen/CFA (instead of collagen/IFA). The graph below presents a comparison between CFA and IFA-containing boosters:

Tail markings and injection location
The needle will be inserted 25 mm from the base of the tail.
If using tail marking for mouse identification, avoid putting marks at that location – markings can make it difficult to see tail veins.
